ELISA TMB Substrate (Sensitive) / Ready-to-use TMB-Substrate is the most common chromogenic substrate for HRP. Horseradish peroxidase (HRP) break down Peroxide and make oxidation on 3,3′,5,5′-tetramethylbenzidine to produce blue colour. After adding Stop solution TMB colour change to yellow which is measurable at 450nm.

Among the existing benzidine substrates that are used for peroxidase-based assays, TMB exhibited the highest sensitivity, better color purity of colored products, and reasonable stability of oxidation products. Ready-to-use TMB (one-component TMB) is a sensitive product of TMB to contamination from a variety of oxidizing agents.

To avoid contamination and premature expiration, avoid contacting TMB solution with any potential source of contamination. Never pipette directly from the bottle. Pour out required amount onto a tube and pipette from the tube. Do not return excess TMB to primary storage container and keep TMB-Substrate solution in dark and refrigerated. One-component TMB is also very sensitive to high temperature and air oxidation, keep main bottle capped to decrease air oxidation and just bring daily TMB-Substrate usage out of main bottle.
