3,3′,5,5′-Tetramethylbenzidine or TMB is a insoluble derivative of Benzidine that after solubilization is used as  chromogenic reagent for  enzyme-linked immunosorbent assays (ELISA). TMB acts as a electron donor for the reduction of  Peroxides to water by peroxidase enzymes. Horseradish peroxidase (HRP), that is a metalloenzyme peroxidase is commonly used in ELISA technique as a conjugated Enzyme that hydrolyses peroxidase in final step of ELISA test to make a blue colored diimine-diamine complex that readily change to yellow color after adding stop solution.

It is not known if TMB is carcinogenic, and the evidence is contradictory, But it has shown  that TMB is not mutagenic by the Ames test, and did not induce formation of tumors in rats. However, TMB (3,3′, 5,5′ tetramethylbenzidine dihydrochloride) is a sensitive chromogen and needs to be cautious in storage and use. TMB is a photosensitive product and cannot be mixed with any oxidative chemicals and should be kept out of direct light and high temperature.

Diagnopal TMB should be stored at 2-4°C but you need to bring its temperature to RT before starting your test assay. This ELISA TMB Substrate for HRP can also be stored at room temperature for up to 6 months that is not recommended. Also, you need to discard TMB-Substrate if solution turns blue or turbid.
