TMB Substrate, which is known as 3,3’,5,5’-tetramethylbenzidine, is a chromogenic chemical component of Benzidine that detects horseradish peroxidase (HRP) activity when oxidized with hydrogen peroxide. It yields a blue color with maximum absorbances at 370 and 652nm. This reagent is supplied as a ready to use component in a mildly acidic solution with a PH of about 3.4. TMB substrate solution provides high signal and high sensitivity and is Recommended for peroxidase-based enzyme immunoassays (ELISA) which has been shown to be a safe-sensitive substrate.

Well stored and unreacted substrate should be colorless or very light-yellow background in appearance. Upon starting the reaction, it develops a deep blue color. The reaction can be stopped using an appropriate stop solution that will produce a soluble yellow dye with the absorbance at 450nm. After stopping the reaction, it is better to read O.D as soon as possible and less than 10 Min.

Diagnopal TMB substrates are available in 3 versions of ready to use solution and can be supplied in a Two-component at your request. Normally, Two- component TMB substrate is more stable than ready to use TMB.
